چکیده

Fibroblasts like synoviocytes (FLS) play several significant roles in rheumatoid arthritis (RA) pathophysiology. This chapter will describe known of FLS disease initiation, joint inflammation, persistence and destruction. It the newly characterized subsets based on single cell RNA sequencing studies, their association to specific aspects disease. Finally, we discuss future targeting treatment RA. The synovial lining layer are identified by surface complement decay-accelerating factor (CD55) along with lubricin metallopeptidase expression. Pathological activation this subset result bone cartilage damage mice. sublining often THY1 expression, but recent studies have highlighted a heterogeneity where distinct additional markers. Sublining expressing human leukocyte antigen-DRA (HLA-DRA) produce C-X-C motif chemokine 12 (CXCL12) receptor activator nuclear factor-κB ligand (RANKL) seems constitute pro-inflammatory that is associated inflammation tertiary lymphoid structures. Another CD34 expression may discriminate common progenitor fibroblast subset. Taken together, isolating characterizing gene report both associations unknown importance markers impose protective or destructive features. supports evidence as active players RA pathology capable cellular recruitment, local crosstalk promotion These discoveries serve an atlas for potential development targeted treatment.

Hypericin-photodynamic therapy inhibits proliferation and induces apoptosis in human rheumatoid arthritis fibroblast-like synoviocytes cell line MH7A

Objective(s): To elucidate the effects and potential mechanisms of hypericin-photodynamic therapy (HYP-PDT) for treating the human rheumatoid arthritis (RA) fibroblast-like synoviocyte (FLS) MH7A cell-line. Materials and Methods: MH7A cells were subjected to HYP-PDT intervention and apoptosis was evaluated via MTT, nuclear staining, and flowcytometry analyses. Intracellular reactive oxygen spec...

BRAF drives synovial fibroblast transformation in rheumatoid arthritis.

Synovial fibroblasts destroy articular cartilage and bone in rheumatoid arthritis, but the mechanism of fibroblast transformation remains elusive. Because gain-of-function mutations of BRAF can transform fibroblasts, we examined BRAF in rheumatoid synovial fibroblasts. The strong gain-of-function mutation, V600R, of BRAF found in melanomas and other cancers was identified in first passage synov...

Interleukin-34 produced by human fibroblast-like synovial cells in rheumatoid arthritis supports osteoclastogenesis

INTRODUCTION Interleukin-34 (IL-34) is a recently defined cytokine, showing a functional overlap with macrophage colony stimulating factor (M-CSF). This study was undertaken to address the expression of IL-34 in rheumatoid arthritis (RA) patients and to investigate its regulation and pathogenic role in RA. METHODS IL-34 levels were determined in the RA synovium, synovial fluid (SF) and fibrob...

Fibroblast-Like Synovial Cells in Rheumatoid Arthritis--the Impact of Infliximab on Hexosaminidase Activity.

BACKGROUND The effect of multiple infusions of infliximab (INF), a chimeric anti-tumor necrosis factor alpha antibody, on the concentration of hexosaminidase (HEX) activity in a synovial cell culture derived from human synovial inflamed fluid obtained from patients suffering from rheumatoid arthritis (RA) has been evaluated. OBJECTIVES The aim of this study was to prove INF efficacy in RA. ...

Profiling drugs for rheumatoid arthritis that inhibit synovial fibroblast activation

Activation of synovial fibroblasts (SFs) contributes to rheumatoid arthritis (RA) by damaging synovial membranes and generating inflammatory cytokines that recruit immune cells to the joint. In this paper we profile cytokine secretion by primary human SFs from healthy tissues and from donors with RA and show that SF activation by TNF, IL-1α, and polyinosinic-polycytidylic acid (Poly(I:C)) cause...
